Stocks began the session in lackluster fashion as the averages opened just above yesterday’s close. There was little early reaction to the morning’s economic reports including the jobless claims data, Philadelphia Fed report, leading economic indicators, and natural gas storage change report. But by mid-day, the Dow had asserted itself as the day’s leader while the Nasdaq struggled to stain positive territory. As the afternoon progressed the Dow continued its advance, the Nasdaq hit 14-year highs, and the S&P also touched a new all-time high. ECONOMIC EVENTS: Jobless Claims data continues to trend lower, with the week of August 16 coming in at 298K vs. a consensus of 300K... The PMI Manufacturing Index Flash was strong at 58.0 for August, up 1.7 points from the July flash of 56.3. COMPANY NEWS: eBay (EBAY) rose almost 5% after a report said that a PayPal spin-off was under consideration... Hormel (HRL) gained almost 5% and hit a 52-week high after reporting better than expected earnings before the market opened... Sears Holding (SHLD) fell over 7% after reporting Q2 earnings this morning that missed on the top and bottom lines. INDEXES: The Dow closed up 60.36, or 0.36%, to 17,039.49, the Nasdaq closed up 5.62, or 0.12%, to 4,532.10, and the S&P 500 closed up 5.86, or 0.29%%, to 1,992.37.

Under the new EMSA, Sucampo receives a new, lower price for certain components of or finished product of lubiprostone globally, except for the U.S. and Canada until the collaboration and license agreement with Takeda Pharmaceutical Company (TKPYY), expires in December 31, 2020, and except for Japan until the manufacturing and supply agreement for Japan expires or certain material circumstances occur. Additionally, under the EMSA Sucampo has the right to qualify a back-up supplier for lubiprostone in expanded circumstances, such as an authorized generic, additional formulations, and/or inability to supply product in certain circumstances, among others. The new EMSA supersedes the current manufacturing and supply agreements with R-Tech except in those situations noted above and is effective as of January 1.

Nektar Therapeutics (NKTR) reports Baxter International (BAX) announced positive results from its Phase 3 pivotal clinical trial of BAX 855, an investigational, extended half-life recombinant factor VIII treatment for hemophilia A based on ADVATE, which met its primary endpoint in reducing annualized bleed rates in the prophylaxis arm compared to the on-demand arm. Top-line results from the prospective, global, multi-center Phase 3 study demonstrated that BAX 855 met its primary endpoint in the control and prevention of bleeding, routine prophylaxis and perioperative management for patients who were 12 years or older. Patients in a twice-weekly prophylaxis arm experienced a 95 percent reduction in median ABR as compared to those in the on-demand arm (1.9 vs. 41.5, respectively). BAX 855 was also effective in treating bleeding episodes, 96 percent of which were controlled with one or two infusions. The half-life of BAX 855 was 1.4 - 1.5 times that of ADVATE, consistent with the findings from the Phase 1 study. No patients developed inhibitors to BAX 855 and no treatment-related serious adverse events, including hypersensitivity, were reported. The most common product-related adverse event was headache. Baxter expects to submit a Biologics License Application for BAX 855 to the U.S. Food and Drug Administration before the end of 2014 and will present additional data in the coming months. In addition to an ongoing continuation study for patients who have completed the pivotal trial, the company is initiating a Phase 3, prospective, open-label, multi-center study to evaluate the safety and efficacy of BAX 855 among 60 previously treated patients under the age of 12 with severe hemophilia A. Consistent with guidelines published by the European Medicines Agency that require a study in children less than 12 years of age prior to filing, Baxter expects to file a Marketing Authorization Application with the EMA upon the completion of the pediatric study

Yet, the ISM-adjusted Philly Fed measure fell to 55.5 in August, after a July surge to a 59.5 cycle-high from 54.1 in June, versus a 49.0 nine-month low in February. The divergence reflected big declines in new and unfilled orders as well as shipments and deliveries that left a more mixed report than implied by the headline surge. The big July vehicle assembly rate pop, and likely ensuing August drop-back, likely fed both the producer sentiment upswing in July and divergent August moves, both within today's report and relative to last Friday's Empire State drop to 14.69 after surging to 24.60 in July, as well as a continued tightening in initial claims. Analysts expect an unchanged Richmond Fed at 4.0, a Dallas Fed up-tick to 13.0 from 12.7, an ISM drop to 55.0 from 57.1, and an ISM-NMI decline to 56.5 from 58.7. The mix should allow the ISM-adjusted average to slip to 55 in August from a 56 cycle-high in July that was also seen in February and March of 2011, versus 54 through Q2 and 52 through Q1.

However, the components were mixed and belie the headline strength. The employment index slid to 9.1 from 12.2 (the latter was the best since October 2013). The workweek edged up to 13.3 from 12.5. New orders were more than halved to 14.7 from 34.2. Prices paid dropped to 24.9 from 34.7, while prices received crumbled to 4.2 from 16.8. The 6-month business conditions index surged to 66.4, however, from 58.1, with employment at 24.7 from 29.1, and capital expenditures at 17.5 from 18.9.

Baxter announced "positive" results from its Phase 3 pivotal clinical trial of BAX 855, which met its primary endpoint in reducing annualized bleed rates in the prophylaxis arm compared to the on-demand arm. Top-line results from the prospective, global, multi-center Phase 3 study demonstrated that BAX 855 met its primary endpoint in the control and prevention of bleeding, routine prophylaxis and perioperative management for patients who were 12 years or older. Patients in a twice-weekly prophylaxis arm experienced a 95% reduction in median ABR as compared to those in the on-demand arm. BAX 855 was also effective in treating bleeding episodes, 96% of which were controlled with one or two infusions. The half-life of BAX 855 was 1.4 – 1.5 times that of ADVATE, consistent with the findings from the Phase 1 study. No patients developed inhibitors to BAX 855 and no treatment-related serious adverse events, including hypersensitivity, were reported. The most common product-related adverse event was headache. Baxter expects to submit a Biologics License Application for BAX 855 to the U.S. Food and Drug Administration before the end of 2014 and will present additional data in the coming months. In addition to an ongoing continuation study for patients who have completed the pivotal trial, the company is initiating a Phase 3, prospective, open-label, multi-center study to evaluate the safety and efficacy of BAX 855 among 60 previously treated patients under the age of 12 with severe hemophilia A. Consistent with guidelines published by the European Medicines Agency that require a study in children less than 12 years of age prior to filing, Baxter expects to file a Marketing Authorization Application with the EMA upon the completion of the pediatric study.

Eli Lilly announces that its investigational medicine ixekizumab was statistically superior to etanercept and placebo on all skin clearance measures in Phase 3 studies. For comparison, between 5 to 7 percent of patients treated with etanercept in the UNCOVER-2 and 3 studies achieved PASI 100. Adverse events were comparable for patients receiving ixekizumab in the 12-week, randomized control portion across all three studies.

Illumina (ILMN) announced it has formed collaborative partnerships with leading pharmaceutical companies to develop a universal next-generation sequencing, or NGS,-based oncology test system. The system will be used for clinical trials of targeted cancer therapies with a goal of developing and commercializing a multi-gene panel for therapeutic selection, resulting in a more comprehensive tool for precision medicine. Initial strategic partners include AstraZeneca (AZN), Janssen Biotech, a Johnson & Johnson (JNJ) company, and Sanofi (SNY). Illumina is working with the strategic partners to develop assays that detect and measure multiple variants simultaneously to support partners’ clinical trials, with the objectives of securing regulatory agency approvals and test commercialization. In parallel, Illumina is working with key thought leaders to set standards for NGS-based assays in routine clinical oncology practice, as well as to define regulatory frameworks to enable this new testing paradigm. Together, Illumina and its strategic partners aim to transition from single-analyte companion diagnostics to panel-based assays that select for “companion therapeutics.”
